细胞简介:
大鼠肝枯否细胞分离自肝脏组织;肝脏是身体内以代谢功能为主的一个器官,并在身体里面起着去氧化、储存肝糖、分泌性蛋白质的合成等作用;肝脏也制造消化系统中之胆汁。肝脏是机体内脏里最大的器官,位于机体中的腹部位置,在右侧横隔膜之下,位于胆囊之前端且于右边肾脏的前方,胃的上方。肝脏是机体消化系统中最大的消化腺,为一红棕色的V字形器官。肝脏是尿素合成的主要器官,又是新陈代谢的重要器官。肝脏在机体位置和形态结构:肝脏位于右上腹,隐藏在右侧膈下和肋骨深面,大部分肝为肋弓所复盖,仅在腹上区、右肋弓间露出并直接接触腹前壁,肝上面则与膈及腹前壁相接。枯否氏细胞(Kupffer细胞)是肝脏的肝血窦内一些固定于窦壁的巨噬细胞,它能吞噬和清除大部分从肠道来的抗原微粒,并能吞噬和清除肝血窦中的细菌、异物和衰老的红细胞,并把血红蛋白分解成胆红素。此外,肝巨噬细胞也有处理抗原,诱导T细胞增殖,参与免疫调节的作用。枯否氏细胞和一般巨噬细胞不同,枯否氏细胞不具有增加抗原免疫原性的能力,相反有消除或减弱抗原性的作用。枯否氏细胞能吞噬来自血液循环的抗原抗体复合物和其他有害物质,以消除这些物质对机体的损害。枯否细胞功能障碍可导致肠源性内毒素血症。电镜下有很多皱褶和微绒毛。

方法简介:
普诺赛实验室分离的大鼠肝枯否细胞采用混合酶灌流消化、低速离心、密度梯度离心、差速贴壁法制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的大鼠肝枯否细胞经CD68免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-R132 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 梭形、巨噬细胞 传代特性 属于终末分化细胞;属于不增殖细胞群 消化液 利多卡因(12mM) 大鼠肝枯否细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
