细胞简介:
大鼠海绵体内皮细胞分离自海绵体组织;阴茎海绵体是由海绵状的小梁和腔隙构成的血窦结构,它主要包括海绵体内皮细胞、平滑肌细胞和成纤维细胞3种细胞成分。其中平滑肌细胞和成纤维细胞镶嵌于海绵体细胞外基质的胶原中,而海绵体内皮细胞则覆盖于海绵体血窦的腔面,仅占海绵体组织的2.8%。在消化海绵体组织时,胶原酶的作用主要是松解海绵体内皮细胞与基膜的联系,破坏海绵体内皮细胞间的紧密连接。如果消化时间延长或胶原酶浓度过大,平滑肌细胞和成纤维细胞也将被消化下来,从而影响海绵体内皮细胞的纯度。因此,比较筛选合适的胶原酶浓度和消化时间是成功分离海绵体内皮细胞的关键。

方法简介:
普诺赛实验室分离的大鼠海绵体内皮细胞采用混合酶消化法结合机械分离法、并用内皮细胞专用培养基培养筛选制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的大鼠海绵体内皮细胞经CD31免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 PLL(0.1mg/ml)或明胶(0.1%) 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-R133 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 内皮细胞样 传代特性 可传2-3代 消化液 0.25%胰蛋白酶 大鼠海绵体内皮细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
