细胞简介:
小鼠神经干细胞分离自脑皮层组织;大脑分左右两个半球,大脑皮质(灰质)覆盖着每个大脑半球的大部分,它是神经元胞体集中的地方。内部则是由神经纤维或髓鞘构成的白质。每一个半球都有三个面,即外侧面(约占整个皮质面积的1/3)、内侧面和底面(占2/3的面积);半球表面有很多深浅不等的沟或裂,沟或裂之间的隆起叫回,它们大大增加了大脑的表面积;大脑外侧面重要的沟、裂有大脑外侧裂、顶枕裂和中央沟。由于三沟裂之界隔,使大脑皮质组分为额叶、顶叶、颞叶、枕叶四大部分。神经干细胞具有分化为神经神经元、星形胶质细胞和少突胶质细胞的能力,能自我更新,并足以提供大量脑组织细胞的细胞群。神经干细胞是一类具有分裂潜能和自更新能力的母细胞,它可以通过不对等的分裂方式产生神经组织的各类细胞。需要强调的是,在脑脊髓等所有神经组织中,不同的神经干细胞类型产生的子代细胞种类不同,分布也不同。神经干细胞作为干细胞的一种,它具有其它所有干细胞的基本特征:具有自我维持和自我更新能力,具有多种分化潜能,具有分化为本系统大部分类型细胞的能力,这种自我更新和分化潜能可以维持相当长的时间甚至终生,对损伤和疾病具有反应能力。神经干细胞在疾病、损伤状态下具有增殖、迁移,并向神经细胞、星形胶质细胞和少突胶质细胞分化的能力,已成为神经系统损伤修复和再生研究的热点,体外建立稳定的神经干细胞培养模型是其基础和临床应用的前提。神经干细胞在培养3-4d后,可形成神经球,神经球在培养基中呈悬浮生长,予以更换半量培基,1周后用吸管轻柔吹打球形克隆成为小的神经球和单细胞悬液,将部分细胞接种到新的培养瓶中,2×10⁵个细胞/瓶,每7-10d传代1次,每隔3d离心更换半量培养基1次,培养条件不变。

方法简介:
普诺赛实验室分离的小鼠神经干细胞采用胰蛋白酶消化后差速贴壁,结合神经干细胞专用培养基培养筛选制备而来,总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的小鼠神经干细胞经Nestin免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 培养基 含B-27 Supplement、EGF、bFGF、Penicillin、Streptomycin等 产品货号 CM-M139 换液频率 每2-3天换液一次 生长特性 悬浮 细胞形态 球形 传代特性 可传2-3代 消化液 0.25%胰蛋白酶,Accutase 小鼠神经干细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
