细胞简介:
小鼠心肌细胞分离自心脏组织;心脏是脊椎动物身体中最重要的一个器官,主要功能是为血液流动提供压力,把血液运行至身体各个部分。心脏由心肌构成,左心房、左心室、右心房、右心室四个腔组成。左右心房之间和左右心室之间均由间隔隔开,故互不相通,心房与心室之间有瓣膜(房室瓣),这些瓣膜使血液只能由心房流入心室,而不能倒流。心脏的作用是推动血液流动,向器官、组织提供充足的血流量,以供应氧和各种营养物质,并带走代谢的终产物(如二氧化碳、无机盐、尿素和尿酸等),使细胞维持正常的代谢和功能。心肌细胞呈菱形、多边形等不规则形状;细胞培养2h后开始贴壁,呈梭形;到12h左右,细胞开始伸出伪足,呈菱形、多角形;细胞分离培养48h以后,大部分伸出伪足、呈巴掌状,部分心肌细胞会出现搏动;心肌细胞为终末分化细胞,在体外不增殖。体外培养的心肌细胞可保持结构及功能上的某些特点,并具有自发性节律搏动,且心肌细胞的培养具有简便、定量、重复性好以及不受神经体液因素的影响等特点。利用培养的心肌细胞在探索非血液动力学因素所致的心肌肥厚的调节,研究心肌细胞的生物力学、凋亡、受体下调、缺血预处理、信号通路、对作用于心脏的新药进行筛选并对其安全性进行评价以及从培养的心肌细胞中提取有价值的生物因子等方面具有广阔的应用前景,对于研究其生理功能、药物作用以及各种致病因素作用下的病理生理改变具重要意义。

方法简介:
普诺赛实验室分离的小鼠心肌细胞采用胶原酶-胰酶联合消化法结合差速贴壁法,并用化学试剂抑制法筛选制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的小鼠心肌细胞经α-Sarcomeric actin免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 PLL(0.1mg/ml) 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-M073 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 梭形、多角形 传代特性 属于终末分化细胞;属于不增殖细胞群 消化液 0.25%胰蛋白酶 小鼠心肌细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
