产品概述
William's E培养基是由William和Gunn设计出来的,主要用于大鼠肝上皮细胞的长期培养,也可用于其他哺乳动物的肝细胞培养。
L-丙氨酰-L-谷氨酰胺(Alanyl-glutamine,Ala-Glu),又名丙氨酰谷氨酰胺、丙谷二肽,是一种高级细胞培养添加剂,可直接替代细胞培养基中的L-谷氨酰胺。L-谷氨酰胺(Glutamine)是细胞培养中所必需的一种营养素,但其在溶液中不稳定,会自发降解生成氨和焦谷氨酸,其中氨对细胞有害;而L-丙氨酰-L-谷氨酰胺在水溶液中十分的稳定,不会自发的降解。细胞利用其机制是:在细胞培养时,细胞会逐渐向培养液中释放一种肽酶,将L-丙氨酰-L-谷氨酰水解成L-丙氨酸和L-谷氨酰胺,而后细胞会将这两种水解产物吸收利用。细胞利用L-丙氨酰-L-谷氨酰的过程与流加培养策略相似,连续的将低浓度水平的L-谷氨酰胺加入到培养液中,从而提高了L-谷氨酰胺的利用率,且不会生成多余的氨,更利于细胞的生长。L-丙氨酰-L-谷氨酰可以代替等摩尔的L-谷氨酰胺,适用于所有的细胞,几乎无需适应,并且可以延长细胞的培养时间,减少传代次数,即节省了时间也节约了金钱。与添加 L-谷氨酰胺的培养基中培养的细胞相比,活性降低得更慢。 延滞期略微延长的原因是肽酶的释放和二肽的消化需要一定的时间。
本产品含有多类细胞培养所需的氨基酸、维生素、无机盐等多种成分,但不含蛋白质、脂类或任何生长因子,故此产品需搭配血清或无血清添加物使用。
