细胞简介:
小鼠脑微血管内皮细胞分离自脑组织;脑微血管内皮细胞是血脑屏障的主要组成成分,它是组成脑微血管腔面单层扁平上皮样细胞,能够限制可溶性物质和细胞等从血液进入大脑,大脑微血管内皮细胞与外周内皮细胞相比具有一些相同特性。它所产生和分泌的生物活性物质对维持血管张力、调节血压、抗血栓形成等有重要作用,在脑血管疾病的发病机制中有重要病理生理学意义。与外周内皮细胞相同,大脑微血管内皮细胞表面表达细胞粘附分子,调控白细胞进入大脑。由于微血管内皮细胞的器官特异性,内皮细胞通常取源于疾病研究的相关组织。其主要特征如下:①脑微血管内皮细胞存在许多细胞间紧密连接,产生很高的跨内皮阻抗,延迟细胞旁的通量;②脑微血管内皮细胞缺乏内皮细胞的窗孔结构,其液相物质胞饮水平较低;③脑微血管内皮细胞具有不对称定位酶和载体介导转运系统,从而产生“两极分化”的表现型。

方法简介:
普诺赛实验室分离的小鼠脑微血管内皮细胞采用胶原酶-中性蛋白酶混合消化法结合密度梯度离心法、最后通过内皮细胞专用培养基培养筛选制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的小鼠脑微血管内皮细胞经CD31免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 PLL(0.1mg/ml)或明胶(0.1%) 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-M108 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 内皮细胞样 传代特性 可传1-2代;不建议多次传代 消化液 0.25%胰蛋白酶 小鼠脑微血管内皮细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
