细胞简介:
小鼠视网膜Muller细胞分离自视网膜组织;视网膜居于眼球壁的内层,是一层透明的薄膜。视网膜由色素上皮层和视网膜感觉层组成,两层间在病理情况下可分开,称为视网膜脱离。色素上皮层与脉络膜紧密相连,由色素上皮细胞组成,它们具有支持和营养光感受器细胞、遮光、散热以及再生和修复等作用。组织学上视网膜分为10层,由外向内分别为:色素上皮层、视锥、视杆细胞层、外界膜、外颗粒层、外丛状层、内颗粒层、内丛状层、神经节细胞层、神经纤维层、内界膜。视网膜内层为衬于血管膜内面的一层薄膜,有感光作用;后部鼻侧有一视神经乳头。视网膜上的感觉层是由三个神经元组成。第一神经元是视细胞层,专司感光,它包括锥细胞和杆细胞。视杆细胞主要在离中心凹较远的视网膜上,而视锥细胞则在中心凹处最多。第二层叫双节细胞,约有10到数百个视细胞通过双节细胞与一个神经节细胞相联系,负责联络作用。第三层叫节细胞层,专管传导。视网膜是一层菲薄的但又非常复杂的结构,它贴于眼球的后壁部,传递来自视网膜感受器冲动的神经纤维跨越视网膜表面,经由视神经到达出口。视网膜的分辨力是不均匀的,在黄斑区,其分辨能力最强。视网膜Muller细胞作为视网膜中的主要胶质细胞,大约占视网膜胶质细胞的90%,因而在视网膜疾病中起着何种作用也受到越来越多的关注。在超微结构水平上,Muller细胞的胞质似乎比临近的其他细胞更高的电子密度,更发达的内质网,细胞核是典型的卵圆形或多角形。但在不同物种中或同一物种中的不同部位,Muller细胞形态也会有所差异的。视网膜Muller细胞是一种特化的神经胶质细胞,不仅具有维持视网膜的正常结构和功能的作用,并调制视网膜神经元活动,还能参与多种病理过程,尤其是眼底增殖性病变,如增生性玻璃体视网膜病变、增生性糖尿病视网膜病变等,体外培养Muller细胞是研究这些增殖性病变途径之一。

方法简介:
普诺赛实验室分离的小鼠视网膜Muller细胞采用胶原酶消化法和胰蛋白酶反复消化法制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的小鼠视网膜Muller细胞经GS免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

包被条件 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-M117 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 梭形、多角形 传代特性 可传3代左右 消化液 0.25%胰蛋白酶 小鼠视网膜Muller细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
