细胞简介:
人胰腺星状细胞分离自胰腺组织;胰腺分为外分泌腺和内分泌腺两部分。外分泌腺由腺泡和腺管组成,腺泡分泌胰液,腺管是胰液排出的通道。胰液中含有碳酸氢钠、胰蛋白酶原、脂肪酶、淀粉酶等。胰液通过胰腺管排入十二指肠,有消化蛋白质、脂肪和糖的作用。内分泌腺由大小不同的细胞团──胰岛所组成,胰岛主要由4种细胞组成:α细胞、β细胞、γ细胞及PP细胞。α细胞分泌胰高血糖素,升高血糖;β细胞分泌胰岛素,降低血糖;γ细胞分泌生长抑素,以旁分泌的方式抑制α、β细胞的分泌;PP细胞分泌胰多肽,抑制胃肠运动、胰液分泌和胆囊收缩。纤维化是慢性胰腺炎的典型病理特征,活化的胰腺星状细胞(PSC)是胰腺纤维化的主要效应细胞,PSC分离和成功培养是体外研究胰腺纤维化的重要前提。未活性化的PSC胞浆中富含维A脂滴,并表达Desmin蛋白,活化后的PSC则表达α-平滑肌肌动蛋白(alpha-SMA)。PSC具有静息态与激活态两种,并分别具有特异的标志物。静息状态PSC胞质内富含的维生素A脂滴可以被油红O染成红色,阳性表达Desmin。激活状态PSC阳性表达alpha-SMA。油红O染色发现,原代分离的细胞培养6d,胞浆中仍可见明显的脂滴,传代后,橙红色的脂滴颗粒显著减少甚至消失。免疫细胞化学染色显示,细胞接种24h仍然表达Desmin,48h后Desmin基本不再表达。培养48h后绝大多数细胞开始表达alpha-SMA,随着培养时间的增长和传代次数的增加,细胞表达alpha-SMA,而不再表达Desmin,说明细胞活化。提示PSC接种24h后即启动了激活过程,至培养第6d大多数细胞激活,传代后细胞处于高度激活状态。原代胰腺星状细胞可作为慢性胰腺炎新药的细胞筛选模型,目前研究发现:胰腺受损时,在各种刺激因子作用下使胰腺星状细胞活化,导致细胞形态、功能发生变化,促使基质增生、胶原蛋白的大量生成及不规则沉积。

方法简介:
普诺赛实验室分离的人胰腺星状细胞采用胶原酶制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的人胰腺星状细胞经α-SMA或Desmin免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

包被条件 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-H024 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 成纤维细胞样 传代特性 可传2-3代 消化液 0.25%胰蛋白酶 人胰腺星状细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
