细胞简介:
人肝内胆管上皮细胞分离自肝脏组织;肝脏是身体内以代谢功能为主的一个器官,并在身体里面起着去氧化、储存肝糖、分泌性蛋白质的合成等作用;肝脏也制造消化系统中之胆汁。肝脏是机体内脏里最大的器官,位于机体中的腹部位置,在右侧横隔膜之下,位于胆囊之前端且于右边肾脏的前方,胃的上方。肝脏是机体消化系统中最大的消化腺,为一红棕色的V字形器官。肝脏是尿素合成的主要器官,又是新陈代谢的重要器官。肝脏在机体位置和形态结构:肝脏位于右上腹,隐藏在右侧膈下和肋骨深面,大部分肝为肋弓所复盖,仅在腹上区、右肋弓间露出并直接接触腹前壁,肝上面则与膈及腹前壁相接。通过控制激素调控的分泌和吸收,在保持、调整和扩大胆小管的结构中发挥重要作用,肝内胆管上皮细胞在先天性和获得性免疫应答方面起着积极作用。肝内胆管上皮细胞约占肝细胞总数的5%,其在肝内胆道系统内形成复杂的网状管形结构。肝内胆管上皮细胞具有复杂的生理代谢功能,参与肝脏的代谢、排泌、免疫等生理过程,在肝脏疾病的发生发展过程中起一定的作用。研究表明,常见的肝内胆管病变例如原发性硬化性胆管炎、胆管癌等疾病,都是以肝内胆管上皮细胞为病变靶位,进而引起肝内胆管上皮损伤。因此,了解正常肝内胆管上皮细胞的生物学特征和病变肝内胆管上皮细胞的病理特征对研究肝内胆管病变的发病机制、诊断和治疗具有重要意义。

方法简介:
普诺赛实验室分离的人肝内胆管上皮细胞采用胶原酶灌注消化法后,再用胶原酶-DNA酶联合消化,接种至预先包被鼠尾胶原的培养瓶中,通过上皮细胞专用培养基培养筛选制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的人肝内胆管上皮细胞经Cytokeratin-19免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 鼠尾胶原Ⅰ(2-5μg/cm2) 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-H042 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 上皮细胞样 传代特性 可传2-3代 消化液 0.25%胰蛋白酶 人肝内胆管上皮细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
