细胞简介:
小鼠肾小管上皮细胞分离自肾组织;肾脏是机体的重要器官,它的基本功能是生成尿液,借以清除体内代谢产物及某些废物、毒物,同时经重吸收功能保留水份及其他有用物质,如葡萄糖、蛋白质、氨基酸、钠离子、钾离子、碳酸氢钠等,以调节水、电解质平衡及维护酸碱平衡。肾脏同时还有内分泌功能,生成肾素、促红细胞生成素、活性维生素D3、前列腺素、激肽等,又为机体部分内分泌激素的降解场所和肾外激素的靶器官。肾脏的这些功能,保证了机体内环境的稳定,使新陈代谢得以正常进行。肾小管是机体肾脏器官上与肾小囊壁层相连的一条细长上皮性小管,具有重吸收(reabsorption)和排泌作用。肾小管按不同的形态结构、分布位置和功能分成近端小管、髓袢和远端小管三部分;近端小管可分为直部和曲部,曲部也称为近曲小管,位于皮质迷路内,于肾小体附近高度蟠曲;远端小管曲部也称为远曲小管,位于皮质迷路内。肾小管上皮细胞是肾小管外面的一层细胞,肾小管上皮细胞的分离、培养是研究肾脏疾病的一项重要技术,目前该细胞分离方法有酶分离法、化学分离法筛网分离法、显微解剖分离法、流式细胞仪分离法等。肾小管上皮细胞主要功能:①肾小管上皮细胞重吸收原尿中几乎全部的葡萄糖和氨基酸;②排泌非营养物质进入终尿;③细胞能分泌炎症介质如细胞因子和趋化因子,通过产生IL-8或直接趋化白细胞参与急性炎症反应;④移植肾炎症和新月体肾炎中PTEpiC表达IL-2R和MHC-Ⅱ类抗原,这说明肾小管上皮细胞参与肾免疫损伤的发生。随着对肾间质纤维化机制研究的日渐加深,肾小管上皮细胞(RTECs)在其中的作用日益被人们重视。因此,提供良好的肾小管上皮细胞模型,在肾小管间质疾病的发病机制和治疗药物筛选的研究中是非常重要的。

方法简介:
普诺赛实验室分离的小鼠肾小管上皮细胞采用先机械研磨过不锈钢网筛分离得到肾小管、后用胶原酶消化,结合上皮细胞专用培养基培养筛选制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的小鼠肾小管上皮细胞经Cytokeratin-18免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 鼠尾胶原Ⅰ(2-5μg/cm2) 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-M062 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 上皮细胞样 传代特性 可传1-2代 消化液 0.25%胰蛋白酶 小鼠肾小管上皮细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
