  4. 细胞简介:

    人胃黏膜上皮细胞分离自胃黏膜组织;胃黏膜柔软,活体呈橘红色。胃空虚时形成许多皱襞,充盈时变平坦。幽门处的黏膜形成环形皱襞,突向腔内称幽门瓣。胃黏膜可分为三层,即上皮层(单层柱状上皮)、固有层(由结缔组织构成、含有大量的胃腺)和黏膜肌层(为薄层平滑肌、排列成内环外纵)。胃黏膜上皮细胞源自胃黏膜的上皮层,细胞为单层柱状上皮,排列整齐,能分泌黏液覆盖于胃黏膜的表面,防止胃酸和胃蛋白酶对胃黏膜的损害。体外培养胃黏膜上皮细胞为研究细胞功能及致病因子、药物、激素对细胞的损伤、保护和功能调控提供了简单而优越的模型。

  5. 方法简介:

    普诺赛实验室分离的人胃黏膜上皮细胞采用胰蛋白酶-胶原酶混合消化法结合差速贴壁法,并通过上皮细胞专用培养基培养筛选制备而来,细胞总量约为5×10⁵cells/瓶。

  6. 质量检测:

    普诺赛实验室分离的人胃黏膜上皮细胞经Cytokeratin-18免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

  7. 培养信息:

    包被条件鼠尾胶原Ⅰ(2-5μg/cm2)
    培养基含FBS、生长添加剂、Penicillin、Streptomycin等

    换液频率每2-3天换液一次
    生长特性贴壁
    细胞形态上皮细胞样
    传代特性可传2-3代
    消化液0.25%胰蛋白酶

    人胃黏膜上皮细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
