细胞简介:
人肾小球系膜细胞分离自肾小球组织;肾小球是肾元中的用于将血液过滤生成原尿的一团毛细血丛,被鲍氏囊所包裹,是尿液形成的重要构造。血液经由入球小动脉进入肾小球。肾小球内的微血管不像其他微血管,汇流入静脉,而是流入出球小动脉。在肾小球内,微血管受到高压,而加速了超滤作用(hyperfiltration)的进行。微血管中的血液经由超滤作用之后,形成滤液,渗入鲍氏囊内。肾小球与鲍氏囊合称为肾小体,肾小球的过滤速率便称为肾小球过滤率。肾小球系膜是位于肾小球毛细血管袢之间的一种特殊间充质,由系膜细胞和系膜基质组成。肾小球系膜细胞是肾小球内非常活跃的细胞,具有分泌细胞基质、产生细胞因子、吞噬和清除大分子物质及类似平滑肌细胞收缩的功能。肾小球系膜细胞增生和基质的过多产生是多种肾小球肾炎的主要病理表现。肾小球系膜细胞的培养是研究系膜扩张、瘢痕形成和肾小球硬化的理想方法。系膜细胞的主要作用可能有:①收缩作用,入球小动脉和出球动脉的收缩作用受系膜细胞的调节,以影响毛细血管袢的内压和滤过率;②支持作用,它填充于毛细血管袢之间,支持毛细血管的位置;③吞噬作用,能吞噬被阻留在基膜内的大分子物质和蛋白质;④分泌肾素,在肾缺血或免疫复合物沉积时,系膜细胞增生且分泌肾素。

方法简介:
普诺赛实验室分离的人肾小球系膜细胞采用机械研磨法结合不同孔径不锈钢网筛过滤后使用胶原酶消化制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的人肾小球系膜细胞经α-SMA免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 PLL(0.1mg/ml) 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-H067 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 成纤维细胞样 传代特性 可传3代左右 消化液 0.25%胰蛋白酶 人肾小球系膜细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
