细胞简介:
人关节软骨细胞分离自关节软骨组织;关节软骨属于透明软骨,表面光滑、呈淡蓝色、有光泽,它是由一种特殊的叫做致密结缔组织的胶原纤维构成的基本框架,这种框架呈半环形,类似拱形球门,其底端紧紧附着在下面的骨质上,上端朝向关节面,这种结构使关节软骨紧紧与骨结合起来而不会掉下来,同时当受到压力时候,还可以有少许的变形,起到缓冲压力的作用。在这些纤维之间,散在分布着软骨细胞,软骨细胞由浅层向深层逐渐由扁平样至椭圆或圆形的细胞组成,这些软骨细胞维持关节软骨的正常代谢。关节软骨没有神经支配,也没有血管,其营养成分必须从关节液中取得,而其代谢废物也必须排至关节液中,关节软骨的这种营养代谢必须通过关节运动,使关节软骨不断的受到压力刺激才行,所以关节运动对于维持关节软骨的正常结构起重要的作用。关节软骨细胞位于关节软骨陷窝内。幼稚的关节软骨细胞位于关节软骨组织的表层,单个分布、体积较小、呈椭圆形,长轴与关节软骨表面平行,越向深层的关节软骨细胞体积之间增大呈圆形,细胞核圆形或卵圆形、染色浅,细胞质弱嗜碱性,常见数量不一的脂滴。成熟的关节软骨细胞多2-8个成群分布于关节软骨陷窝内,这些关节软骨细胞由同一个母细胞分裂增殖而成,称为同源细胞群。电镜下,关节软骨细胞有突起和皱褶,细胞质内有大量的粗面内质网和发达的高尔基复合体及少量的线粒体。在组织切片中,关节软骨细胞收缩为不规则形,在软骨囊和细胞之间出现较大的腔隙。体外培养的关节软骨细胞对于研究其生理功能、药物作用以及各种致病因素作用下的病理生理改变具重要意义。

方法简介:
普诺赛实验室分离的人关节软骨细胞采用胶原酶联合中性蛋白酶消化制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的人关节软骨细胞经Ⅱ型胶原蛋白免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-H096 换液频率 每3-4天换液一次 生长特性 贴壁 细胞形态 梭形、多角形 传代特性 可传5代左右;3代以内状态最佳 消化液 0.25%胰蛋白酶 人关节软骨细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
