细胞简介:
人支气管上皮细胞分离自支气管组织;支气管(Bronchi),是指由气管分出的各级分枝,由气管分出的一级支气管,即左、右主支气管。左主支气管与右主支气管相比较,前者较细长,走向倾斜;后者较粗短,走向较前者略直,所以经气管堕入的异物多进入右主支气管。支气管和气管还有以区别就是,气管是以“C”型的气管软骨为支架,而支气管不是。支气管上皮是气道与外界环境接触的第一道防线,不仅是各种病原体、炎症介质作用的靶细胞,还作为效应细胞合成、释放多种炎性介质和细胞因子,从而参与气道炎症及免疫反应。支气管上皮细胞在哮喘、慢性阻塞性肺疾病、肺癌、肺囊性纤维化以及一些感染性呼吸道疾病的发病机制中均起着重要的作用。体外培养的原代支气管上皮细胞因与体内组织在形态结构和功能活动上存在很大的相似性,因此,在基础及临床研究中极为重要。

方法简介:
普诺赛实验室分离的人支气管上皮细胞采用链霉蛋白酶-中性蛋白酶混合消化法结合机械刮刷并通过上皮细胞专用培养基培养筛选制备而来,细胞总量约为5×10⁵cells/瓶。

质量检测:
普诺赛实验室分离的人支气管上皮细胞经PCK免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

培养信息:
包被条件 鼠尾胶原Ⅰ(2-5μg/cm2) 培养基 含FBS、生长添加剂、Penicillin、Streptomycin等 产品货号 CM-H009 换液频率 每2-3天换液一次 生长特性 贴壁 细胞形态 上皮细胞样 传代特性 可传2-3代 消化液 0.25%胰蛋白酶 人支气管上皮细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
