  4. 细胞简介:

    人淋巴管内皮细胞分离自淋巴管组织;淋巴管由毛细淋巴管汇合而成。其形态结构与静脉相似,但管径较细,管壁较薄,瓣膜较多且发达,外形呈串珠状。淋巴管根据其位置分为浅、深二种。它们管位于皮下,常与浅静脉伴行,收集皮肤和皮下组织的淋巴。深淋巴管与深部血管伴行,收集肌肉和内脏的淋巴。浅、深淋巴管之间有广泛的交通支。淋巴管在向心行程中,通常经过一个或多个淋巴结,从而把淋巴细胞带入淋巴液。主要功能是滤过淋巴液,产生淋巴细胞和浆细胞,参与机体的免疫反应。当局部感染时,细菌、病毒或癌细胞等可沿淋巴管侵入,引起局部淋巴结肿大。如该淋巴结不能阻止和消灭它们,则病变可沿淋巴管的流注方向扩散和转移。淋巴管内皮细胞(LEC)是衬覆于淋巴管内表面的一种单层扁平上皮,是构成淋巴管壁的主要结构,参与维持体液平衡,调节淋巴细胞再循环和机体的免疫反应和组织液及蛋白质的运输,在疾病过程中也起着重要作用。近年研究表明,LEC还在伤口愈合、淋巴管水肿和炎症扩散等病理过程中起重要作用,而且与肿瘤转移密切相关。淋巴管内皮细胞主要功能:①调节体液、蛋白和组织压力平衡;②为免疫系统的重要组成部分。淋巴管内皮细胞与主要病生理变化:①囊肿型淋巴管瘤;②淋巴管炎;③淋巴结核。

  5. 方法简介:

    普诺赛实验室分离的人淋巴管内皮细胞采用中性蛋白酶消化法结合内皮细胞专用培养基培养筛选制备而来,细胞总量约为5×10⁵cells/瓶。

  6. 质量检测:

    普诺赛实验室分离的人淋巴管内皮细胞经VEGFR3免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

  7. 培养信息:

    包被条件PLL(0.1mg/ml)或明胶(0.1%)
    培养基含FBS、生长添加剂、Penicillin、Streptomycin等

    消化液0.25%胰蛋白酶

    人淋巴管内皮细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
