背景描述 | 293T/17细胞株是293T细胞株的衍生株,293T是293 [HEK-293]细胞株插入了SV40 T-antigen的温度敏感基因形成的高转衍生株。293T细胞进行克隆,检测pBND和pZAP载体,得到一株能生成高滴度感染逆转录病毒的衍生株293T/17细胞。293T/17细胞持续表达猿病毒40(SV40)大T抗原,而因其高转染能力筛选到17号克隆。293T/17细胞用pCRIPenv-和pCRIPgag-2载体转染得到ANJOU65细胞株,ANJOU65细胞再用pCRIPgag-2和pGPT2E载体转染得到亲宅性外壳表达细胞株BOSC 23。ANJOU65细胞用携带gpt抗性基因的pCRIPAMgag载体转染得到Bing双向性表达包装细胞株。 |
