  4. 细胞简介:

    人口腔上皮细胞分离自口腔黏膜组织;口腔上皮细胞是一种上皮细胞,呈扁平、多边形,形状不规则。口腔各壁都有黏膜覆盖,口腔上皮细胞主要分布在口腔两侧颊部。上皮的垂直切面上,细胞形状不一。紧靠基膜的一层基底细胞为矮柱状,为具有增殖分化能力的干细胞,部分子细胞向浅层移动。基底层以上是数层多边形细胞,再上为几层梭形或扁平细胞。仅靠近表面几层细胞为扁平状,基底层细胞能不断分裂增生,以补充表层衰老或损伤脱落的细胞。复层扁平上皮深层的结缔组织内有丰富的毛细血管,有利于复层扁平上皮的营养。

  5. 方法简介:

    普诺赛实验室分离的人口腔上皮细胞采用胰蛋白酶-胶原酶混合消化法结合差速贴壁法,并通过上皮细胞专用培养基培养筛选制备而来,细胞总量约为5×10⁵cells/瓶。

  6. 质量检测:

    普诺赛实验室分离的人口腔上皮细胞经Cytokeratin-18免疫荧光鉴定,纯度可达90%以上,且不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌等。

  7. 培养信息:

    包被条件鼠尾胶原Ⅰ(2-5μg/cm2)
    培养基含FBS、生长添加剂、Penicillin、Streptomycin等
    产品货号CM-H203
    换液频率每2-3天换液一次
    生长特性贴壁
    细胞形态上皮细胞样
    传代特性可传2-3代
    消化液0.25%胰蛋白酶

    人口腔上皮细胞体外培养周期有限;建议使用普诺赛配套的专用生长培养基及正确的操作方法来培养,以此保证该细胞的最佳培养状态。
