产品概述
MTT全称为3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide,汉语化学名为 3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐,商品名:噻唑蓝,是一种黄颜色的染料。MTT能够检测细胞存活和生长,其检测原理为:活细胞线粒体中的琥珀酸脱氢酶,能使外源性MTT还原为不溶于水的蓝紫色结晶物--甲瓒(Formazan),并沉积在细胞中,而死细胞则无此功能;二甲基亚砜(DMSO)能溶解细胞中的甲瓒,用酶标仪在490nm或570nm波长处(常用490nm)测定其光吸收值(OD值),通过OD值的大小,可以判断活细胞的数量或活性,在一定细胞数范围内,MTT结晶形成的量与活细胞的数量或活性成正比。该方法的特点是灵敏度高,并且十分经济,已广泛应用于一些生物活性因子的活性检测、大规模的抗肿瘤药物筛选、细胞毒性试验以及肿瘤放射敏感性测定等。

注意事项
1、本产品经0.22 μm过滤除菌,使用本产品时应注意无菌操作,避免污染。
2、MTT溶液为黄色溶液,见光易分解,操作时应避光,当MTT溶液颜色变为灰绿色时,请勿使用。
3、MTT溶液在4℃、冰浴等较低温度情况下会凝固,可以在20℃-25℃水浴中温育片刻至全部融解混匀后使用。
4、使用MTT时应先混匀,不宜长时间放置于室温环境;2-8℃避光保存时,一个月内有效;切忌反复冻融,用量较少时建议分装冻存。
5、MTT对菌很敏感,培养细胞时尽量细菌避免污染。
6、MTT有致癌性,应做好防护工作。
7、本产品仅用于科研或进一步研究使用,不用于诊断和治疗。
