注意事项 | 1.该细胞并不形成一致的单层,而是形成集落。2.该细胞会使培养基快速变酸,且生长缓慢,故传代后 48 小时内不应扰动;3.普通TC处理的培养瓶或皿不能使细胞很好的贴壁,培养难度较高,需使用 Corning 的 cellbind 细胞培养瓶(货号是 3289),或者使用多聚-L-赖氨酸溶液(货号 PB180523)包被过的培养皿;4.在细胞运输途中,多数细胞会从培养瓶底分离,大片悬浮在培养基中,按照收货注意事项处理即可恢复正常。 |
背景描述 | 人前列腺癌细胞LNCaP clone FGC是从一位50岁白人男性(血型B+)的左锁骨淋巴结针刺活检中分离,该患者经确诊为前列腺癌转移。LNCaP clone FGC细胞对5-α-二氢睾酮(生长调节子和酸性磷酸脂酶产物)有响应。LNCaP clone FGC细胞并不形成一致的单层,而是形成集落,在传代时可以用滴管反复吹吸打碎。LNCaP clone FGC细胞仅仅轻轻地吸附在基底上,不形成汇合,很快使培养基变酸。LNCaP clone FGC细胞生长极其缓慢,传代后48小时内不应扰动。当培养瓶封包后,多数LNCaP clone FGC细胞从培养瓶底分离,悬浮在培养基中。收到后,在通常培养单层细胞的条件下培养24-48小时,以使细胞再贴壁。此后,可以换上新鲜培养液。如果需要,培养瓶内容物可以收集,300g离心15分钟,以10毫升培养液重悬并培养到一个单独的培养瓶中。 |
