注意事项 | 1. SK-BR-3细胞从冷冻保存中恢复的速度可能很慢。SK-BR-3细胞在低温保存恢复过程中的附着会很轻。这很正常。这种细胞系在培养的整个第一周以及每次继代培养后都表现出松散的附着和漂浮的细胞,这并不少见;2. 如果存在大量漂浮物,尤其是在生长的第一周,在启动复苏后几天内保持细胞不受干扰。如果仍然有很多漂浮物,用台盼蓝检查生存能力。通过轻轻离心保留漂浮细胞,并在更换培养基时将其与附着的细胞一起添加回同一培养容器中,这一点很重要。不要分离或丢弃漂浮细胞。细胞最初以小斑块或细胞群的形式附着,许多细胞团仍处于悬浮状态。几天后,生长将从贴壁细胞群向外延伸。通常也会出现一些细胞碎片。漂浮细胞应始终通过温和离心(125xg,持续5-7min)保留,并在换液或传代后放回培养容器;3. SK-BR-3细胞倾向于相互堆积。在细胞达到70-80%汇合度之前不要用胰酶消化处理细胞。如果让细胞过度生长,细胞就会分离漂浮。 |
背景描述 | SK-BR-3 [SKBR3]细胞是由Trempe·G和Old·L·J于1970年从一位43岁的白人女性乳腺癌患者的胸腔积液中分离得到的。SK-BR-3 [SKBR3]细胞亚显微结构特征包括微丝和桥粒、肝糖原颗粒、大溶酶体、成束的细胞质纤丝;SK-BR-3 [SKBR3]细胞过表达HER2/c-erb-2基因产物。 |
